Abstract:
In vivo fluorescent cellular imaging of deep tissue is highly challenging, because the excitation needs to penetrate through strong scattering tissue and the emission signal is degraded significantly by photon diffusion induced by tissue-scattering. To battle the challenge, our group combined Bessel light-sheet microscopy with nonlinear structured illumination microscopy (SIM) to produce cellular-resolution diffusion-free image of deep structures. With two-photon Bessel SIM light sheet excitation, tissue up to 600 microns wide can be imaged with 500 microns penetration depth, and clear images can be obtained at cellular resolution in depth beyond 200 microns, allowing noninvasive studying of deep organ in live embryos. With one-photon excitation, the spectral Bessel SIM light sheet technique can perform hyper-spectral imaging with true parallel detection on multiple excitation channels, which opens the door to fast multiplexed light-sheet imaging in live tissue.
